TAQUITO: A NEW AUTONOMOUS SENSOR FOR FIRST FLASH MECHANICALLY STIMULATED BIOLUMINESCENCE

Conventional flow-through devices measuring bioluminescence emissions of marine plankton generally use an initial stimulation from an impeller pump or flow restriction before subsequent light emissions are quantified in an integrating cavity coupled to a sensitive detector. A commercially available example is the Underwater Bioluminescence Assessment Tool (UBAT). A drawback of this approach is the emission kinetics from organisms in the chamber are occurring in different overlapping phases depending on exactly when they were stimulated, i.e., while some organisms are just starting their emissions others that entered the cavity earlier are finishing their emissions, so resulting output can be difficult to interpret in terms of organism flash kinetics. For example, these sensors will provide a single baseline value with some minor fluctuations for a relatively homogenous assemblage of bioluminescent dinoflagellates with sufficient abundance.

TAQUITO (Tubular Autonomous QUantitative Intensity Tracking Optics) bathyphotometer is being developed to measure first flash kinetics, where organisms are drawn through a tube lined with an array of sensitive SiPM detectors after being stimulated by a grid at the mouth of the tube. The detector array quantifies the intensity of first flash bioluminescence over time after initial stimulation. The design is analogous to the historical HIDEX device deployed by NAVOCEANO from 1995-2005 and the CODEX sensor recently developed at HBOI-FAU, but made much more compact, low power, with a flow rate comparable to UBAT, so the sensor is suitable for autonomous platforms. The mechanical-electrical-optical design and measurements from prototype embodiments will be presented.
