OBSERVATION OF PHYTOPLANKTON COMMUNITIES IN THE SENEGALESE UPWELLING SYSTEM: IN VIVO/IN SITU MULTI-SPECTRAL FLUOROMETRY APPROACH
In vivo/in situ observations have been carried out during different sampling campaigns to study the seasonal, horizontal and vertical distribution of phytoplankton in the Senegalese upwelling system. Phytoplankton has been studied using different methods: multi-spectral fluorometry (FluoroProbe bbe Moldaenke Gmbh) used in continuous subsurface measurements and on water column profiles, automated underway cytometry of the CytoSense/Sub (Cytobuoy) type, discrete pigment concentration sampling (by HPLC: High Performance Liquid Chromatography), taxonomy of the micro-phytoplankton by inverted optical microscopy and DNA sequencing (metabarcoding). The work focuses on a fluorescence spectrum analysis that uses principal component analysis (PCA) coupled with K-means to classify phytoplankton assemblages. Key results feature a large variability of the distribution of phytoplankton assemblages, characterized by their functional group, over the seasons, from the upwelling phase and the coast to the offshore, with consequences on the structure and functioning of ecosystems in southern Senegal, as well as their productivity, and highlights the ability of the method to observe changes in assemblies between the sub-provinces of the system at the regional scale.
